ABSTRACT

Platycodonis Radix is the dry root of Platycodon grandiflorum of Campanulaceae, which has a variety of pharmacological effects and is a commonly used bulk Chinese medicine. In this study, the chloroplast genome sequences of six P. grandiflorum from different producing areas has been sequenced with Illumina HiSeq X Ten platform. The specific DNA barcodes were screened, and the germplasm resources and genetic diversity were analyzed according to the specific barcodes. The total length of the chloroplast genome of 6 P. grandiflorum samples was 172 260-172 275 bp, and all chloroplast genomes showed a typical circular tetrad structure and encoded 141 genes. The comparative genomics analysis and results of amplification efficiency demonstrated that trnG-UCC and ndhG_ndhF were the potential specific DNA barcodes for identification the germplasm resources of P. grandiflorum. A total of 305 P. grandiflorum samples were collected from 15 production areas in 9 provinces, for which the fragments of trnG-UCC and ndhG_ndhF were amplificated and the sequences were analyzed. The results showed that trnG-UCC and ndhG_ndhF have 5 and 11 mutation sites, respectively, and 5 and 7 haplotypes were identified, respectively. The combined analysis of the two sequences formed 13 haplotypes (named Hap1-Hap13), and Hap4 is the main genotype, followed by Hap1. The unique haplotypes possessed by the three producing areas can be used as DNA molecular tags in this area to distinguish from the germplasm resources of P. grandiflorum from other areas. The haplotype diversity, nucleotide diversity and genetic distance were 0.94, 4.79×10-3 and 0.000 0-0.020 3, respectively, suggesting that the genetic diversity was abundant and intraspecific kinship was relatively close. This study laid a foundation for the identification of P. grandiflorum, the protection and utilization of germplasm resources, and molecular breeding.

ABSTRACT

RESUMEN La cebadilla criolla (Bromus catharticus Vahl) es la gramínea forrajera nativa anual/bianual de clima templado de mayor importancia y difusión en la Argentina. Varios autores han estudiado la variabilidad fenotípica en caracteres morfofisiológicos en cultivares y poblaciones nativas de cebadilla criolla. Las primeras variedades cultivadas comerciales de cebadilla criolla, eran poblaciones heterogéneas a las que se las consideraba de mejor adaptación a toda la región de cultivo. Con los estudios de caracterización genética, agronómica y molecular de las poblaciones base de selección y del germoplasma conservado en el banco activo de INTA Pergamino, se contribuyó a conocer y conservar la diversidad genética disponible y al mismo tiempo, disponer de variabilidad genética utilizable en programas de mejoramiento. Los cultivares públicos más representativos considerados en este trabajo fueron: Pergamino Martín Fierro MAG, Fierro Plus INTA, Bar INTA 200, Rosalía INTA e INTA Calvu, en los que se desarrollaron paquetes tecnológicos asociados a la utilización en producción animal.


ABSTRACT The prairie grass (Bromus catharticus Vahl) is the annual/biennial native forage grass of temperate climates of most importance and diffusion in Argentina. Several authors have studied the phenotypic variability in morphophysiological characters in cultivars and native populations of prairie grass. The first commercial cultivars were heterogeneous populations that were considered the best adapted in the temperate region of Argentina. Genetic, agronomic and molecular characterization of the selection base populations and of the germplasm conserved in the active bank of INTA Pergamino, contributed to knowing and conserving the available genetic diversity and, at the same time, provided usable genetic variability in breeding. The most representative cultivars considered in this work were Pergamino Martín Fierro MAG; Fierro Plus INTA, Bar INTA 200, Rosalía INTA and INTA Calvu, in which technological packages associated with the utilization in animal production were developed.

Abstract Allium sativum L. is an herb of the Alliaceae family with a specific taste and aroma and medicinal and nutraceutical properties that are widely marketed in several countries. Brazil is one of the largest importers of garlic in the world, despite of its production is restricted and limited to internal consumption. Thus, explore the genetic diversity of commercial garlic conserved at germplasm banks is essential to generate additional genetic information about its economically important crop. A suitable tool for this purpose is the cytogenetic characterisation of these accessions. This study aimed to characterise the cytogenetic diversity among seven accessions of garlic from a Germplasm Bank in Brazil. The karyotypes were obtained by conventional staining and with chromomycin A3 (CMA) and 4,6-diamidino-2-phenylindole (DAPI) fluorochromes. All accessions analysed showed chromosome number 2n = 16, karyotype formula 6M+2SM, symmetrical karyotypes, reticulate interphase nuclei, and chromosomes with uniform chromatin condensation from prophase to metaphase. The fluorochromes staining showed differences in the amount and distribution of heterochromatin along the chromosomes and between accessions studied. Based on the distribution pattern of these small polymorphisms, it was possible to separate the seven accessions into three groups. It was also possible to differentiate some of the accessions individually. One of the results obtained showed a heteromorphic distension of the nucleolar organiser region observed on the chromosome pairs 6 or 7 with peculiar characteristics. It was suggested for example, that the heteromorphic block of heterochromatin (CMA+++/DAPI-) on chromosome 6 of the "Branco Mineiro Piauí" accession can be used as a marker to identify this genotype or may be associated with some character of economic interest.


Resumo Allium sativum L. é uma erva da família Alliaceae com sabor e aroma específicos e propriedades medicinais e nutracêuticas amplamente comercializada em diversos países. O Brasil é um dos maiores importadores de alho do mundo, apesar da sua produção ser restrita e limitada ao consumo interno. Assim, explorar a diversidade genética do alho comercial conservado em bancos de germoplasma é essencial para fornecer informações genéticas adicionais acerca dessa cultura economicamente importante. Uma ferramenta adequada para esse fim é a caracterização citogenética desses acessos. Este estudo teve como objetivo caracterizar a diversidade citogenética entre sete acessos de alho de um Banco de Germoplasma no Brasil. Os cariótipos foram obtidos por coloração convencional e com os fluorocromos de cromomicina A3 (CMA) e 4,6-diamidino-2-fenilindol (DAPI). Todos os acessos analisados ​​apresentaram número cromossômico 2n = 16, fórmula cariotípica 6M + 2SM, cariótipos simétricos, núcleos reticulados em intérfase e cromossomos com condensação uniforme da cromatina da prófase para a metáfase. ABSTRACT

This study aimed to develop a protocol for the cryopreservation of Pseudoplatystoma corruscans semen. For this, mature males were hormonally induced with a single dose of carp pituitary extract (5 mg/kg body weight). Semen was collected and evaluated. Two cryoprotectants were tested to compose the diluents: dimethyl acetamide (DMA) and dimethyl sulfoxide (Me2SO), in two concentrations (8% and 10%), + 5.0% glucose + 10% egg yolk. The semen was diluted in a 1: 4 ratio (semen: extender), packed in 0.5 mL straws and frozen in a dry shipper container in liquid nitrogen vapors. After thawing, sperm kinetics, sperm morphology and DNA integrity of cryopreserved sperm were evaluated. Pseudoplatystoma corruscans males produced semen with sperm motility > 80%. After thawing, all treatments provided semen with total sperm motility > 40%, with no significant difference (P < 0.05) between them, as well as between the other sperm kinetic parameters evaluated. The treatments with DMA provided a smaller fragmentation of the DNA of the gametes. Sperm malformations were identified in both fresh and cryopreserved semen, with a slight increase in these malformations being identified in sperm from thawed P. corruscans semen samples.(AU)


Este estudo teve como objetivo desenvolver um protocolo para a criopreservação do sêmen de Pseudoplatystoma corruscans. Para tal, machos maduros foram induzidos hormonalmente com uma dose única de extrato de hipófise de carpa (5 mg/kg de peso vivo). O sêmen foi coletado e avaliado. Sendo testados para compor os diluentes, dois crioprotetores: dimetil acetamida (DMA) e dimetil sulfóxido (Me2SO), em duas concentrações (8% e 10%), + 5,0% glicose + 10% gema de ovo. O sêmen foi diluído na proporção 1: 4 (sêmen: extensor), embalado em palhetas de 0,5 mL e congelado em container dryshipper em vapores de nitrogênio líquido. Após o descongelamento, foram avaliados os aspectos cinéticos espermáticos, a morfologia espermática e a integridade do DNA dos espermatozoides criopreservados. Os machos de P. corruscans produziram sêmen com motilidade espermática > 80%. Todos os tratamentos proporcionaram após o descongelamento sêmen com motilidade espermática total > 40%, sem diferença significativa (P < 0,05) entre eles, como também entre os demais parâmetros cinéticos espermáticos avaliados. Os tratamentos com DMA proporcionaram uma menor fragmentação do DNA dos gametas. Malformações espermáticas foram identificadas, tanto no sêmen fresco, como no criopreservado, sendo identificado um aumento discreto dessas malformações nos espermatozoides das amostras de sêmen descongeladas de P. corruscans.(AU)

ABSTRACT

Ganoderma lucidum is a valuable medical macrofungus with a myriad of diverse secondary metabolites, in which triterpenoids are the major constituents. This paper introduced the germplasm resources of genus Ganoderma from textual research, its distribution and identification at the molecular level. Also we overviewed G. lucidum in the components, the biological activities and biosynthetic pathways of ganoderic acid, aiming to provide scientific evidence for the development and utilization of G. lucidum germplasm resources and the biosynthesis of ganoderic acid.

ABSTRACT

Abstract The genus Capsicum, native to tropical and subtropical America, belongs to the Solanaceae family, which includes commercially important vegetables such as chilies and green peppers. The silverleaf whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae), causes losses to vegetables including Capsicum species. Among the alternatives of pest control, an effective, economical, and environmentally compatible method is the resistance of the host plant. Infestation by B. tabaci was evaluated in 73 Capsicum genotypes, corresponding to the species C. annuum, C. baccatum, C. sinense, C. frutescens and C. pubescens from an Ecuadorian genebank. Eighty-four percent of the C. baccatum genotypes evaluated showed the highest population densities of B. tabaci, while all the genotypes of C. sinense and C. frutescens had the lowest values (p < 0.05). The non-preference of adults and the scarce oviposition of B. tabaci on genotypes of C. sinense and C. frutescens suggests resistance due to antixenosis. These results could guide breeding programs for the resistance of Capsicum species to B. tabaci infestations.


Resumen El género Capsicum es nativo de América tropical y subtropical, pertenece a la familia Solanaceae e incluye ajíes y pimientos, que son hortalizas comercialmente importantes. La mosca blanca Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) ocasiona pérdidas en hortalizas incluyendo especies de Capsicum. Entre las alternativas de control de plagas, un método eficaz, económico y ambientemente compatible es la resistencia de la planta hospedera. Se evaluó la infestación por B. tabaci en 73 genotipos de Capsicum nativos de Ecuador, correspondientes a las especies C. annuum, C. baccatum, C. chinense, C. frutescens y C. pubescens, provenientes de un banco de germoplasma ecuatoriano. El 84% de los genotipos de C. baccatum evaluados mostraron las mayores densidades poblacionales de B. tabaci, mientras que los menores valores los tuvieron todos los genotipos de C. sinense y C. frutescens (p < 0.05). La no preferencia de adultos y la escasa oviposición de B. tabaci sobre genotipos de C. sinense y C. frutescens sugiere resistencia por antixenosis. Estos resultados podrían orientar programas de mejoramiento genético para la resistencia de especies de Capsicum ante infestaciones por B. tabaci.

ABSTRACT

Habb-ul-Aas is a commercial drug obtained from Myrtus communis L and widely used for abdominal disorders in Pakistan. This sclerophyllous shrub is commonly found in the Lower Dir, Malakand Division, Pakistan. This plant is used as crude drug in herbal as well as pharmaceutical industry due to presence of aromatic oil obtained from various parts of the plant. Two varieties viz., italic and baetica of the plant exist in the world; however, no study reported before about the existence of this species up to the varietal level from Pakistan. The current study was undertaken to characterize the existing germplasm by using certain morphometric parameters in relation to physiognomic features. They exhibit high variability among characters, especially for characteristics useful in selecting cultivars for commercial production. Among the various ecotypes, MBT3 possessed high shoot length (29.96cm), leaf area (2.3cm), fruit volume (0.97ml) & fruit weight (0.88g) and pulp (0.87g) and therefore recommended for the cultivar development.

ABSTRACT

Selenicereus megalanthus H. is a tropical fruit belonging to the family Cactaceae, is rich in essential nutrients, antioxidants and bioactive components. It presents wide variability in different characteristics and a great demand in the market; however, genetic studies in Colombia are scarce. The main of this study was to characterize the genetic diversity of 76 yellow pitahaya genotypes with eight ISSR markers. Genetic parameters expected average heterozygosity (He), percentage of polymorphic loci, genetic distances and Fst were estimated with TFPGA. The analysis of the population genetic structure was carried out with the STRUCTURE 2.3.4. As a result, 225 alleles were generated and the number of polymorphic loci ranged 85 (CT, AG) to 90 (GT). High genetic diversity was found, with an average value of heterozygosity was 0.34 with a genetic differentiation coefficient (Fst) of 0.26, indicating that there was a great genetic diversity, similar values than those reported in other studies of pitahaya genetic diversity in Colombia. The 76 genotypes were grouped into K=3 according to geographic location, however, in some groups a mixture of individuals from different origins was observed. The analysis of molecular variance (AMOVA) showed higher variation (75%) within groups than among groups (25%). These results provide information that can be used to develop conservation strategies for dragon fruit and breeding programs to obtain more productive pitahaya genotypes with superior quality, high yield and with resistance to biotic and abiotic factors.

Apresenta grande variabilidade em diferentes características e uma grande demanda no mercado; no entanto, os estudos genéticos na Colômbia são escassos. O principal deste estudo foi caracterizar a diversidade genética de 76 genótipos de pitahaya amarela com oito marcadores ISSR. Parâmetros genéticos esperados de heterozigosidade média (He), porcentagem de locos polimórficos, distâncias genéticas e Fst foram estimados com TFPGA. A análise da estrutura genética da população foi realizada com a ESTRUTURA 2.3.4. Como resultado, 225 alelos foram gerados e o número de loci polimórficos variou de 85 (CT, AG) a 90 (GT). Foi encontrada alta diversidade genética, com um valor médio de heterozigosidade de 0,34 com coeficiente de diferenciação genética (Fst) de 0,26, indicando que havia uma grande diversidade genética, valores semelhantes aos relatados em outros estudos de diversidade genética de pitahaya na Colômbia. Os 76 genótipos foram agrupados em K = 3 de acordo com a localização geográfica, porém, em alguns grupos foi observada uma mistura de indivíduos de diferentes origens. A análise de variância molecular (AMOVA) mostrou maior variação (75%) dentro dos grupos do que entre os grupos (25%). Esses resultados fornecem informações que podem ser utilizadas para desenvolver estratégias de conservação da fruta do dragão e programas de melhoramento para a obtenção de genótipos de pitahaya mais produtivos, com qualidade superior, alto rendimento e com resistência a fatores bióticos e abióticos.

Abstract Selenicereus megalanthus H. is a tropical fruit belonging to the family Cactaceae, is rich in essential nutrients, antioxidants and bioactive components. It presents wide variability in different characteristics and a great demand in the market; however, genetic studies in Colombia are scarce. The main of this study was to characterize the genetic diversity of 76 yellow pitahaya genotypes with eight ISSR markers. Genetic parameters expected average heterozygosity (He), percentage of polymorphic loci, genetic distances and Fst were estimated with TFPGA. The analysis of the population genetic structure was carried out with the STRUCTURE 2.3.4. As a result, 225 alleles were generated and the number of polymorphic loci ranged 85 (CT, AG) to 90 (GT). High genetic diversity was found, with an average value of heterozygosity was 0.34 with a genetic differentiation coefficient (Fst) of 0.26, indicating that there was a great genetic diversity, similar values than those reported in other studies of pitahaya genetic diversity in Colombia. The 76 genotypes were grouped into K=3 according to geographic location, however, in some groups a mixture of individuals from different origins was observed. The analysis of molecular variance (AMOVA) showed higher variation (75%) within groups than among groups (25%). These results provide information that can be used to develop conservation strategies for dragon fruit and breeding programs to obtain more productive pitahaya genotypes with superior quality, high yield and with resistance to biotic and abiotic factors.


Resumo Selenicereus megalanthus H. é uma fruta tropical pertencente à família Cactaceae, rica em nutrientes essenciais, antioxidantes e componentes bioativos. Apresenta grande variabilidade em diferentes características e uma grande demanda no mercado; no entanto, os estudos genéticos na Colômbia são escassos. O principal deste estudo foi caracterizar a diversidade genética de 76 genótipos de pitahaya amarela com oito marcadores ISSR. Parâmetros genéticos esperados de heterozigosidade média (He), porcentagem de locos polimórficos, distâncias genéticas e Fst foram estimados com TFPGA. A análise da estrutura genética da população foi realizada com a ESTRUTURA 2.3.4. Como resultado, 225 alelos foram gerados e o número de loci polimórficos variou de 85 (CT, AG) a 90 (GT). Foi encontrada alta diversidade genética, com um valor médio de heterozigosidade de 0,34 com coeficiente de diferenciação genética (Fst) de 0,26, indicando que havia uma grande diversidade genética, valores semelhantes aos relatados em outros estudos de diversidade genética de pitahaya na Colômbia. Os 76 genótipos foram agrupados em K = 3 de acordo com a localização geográfica, porém, em alguns grupos foi observada uma mistura de indivíduos de diferentes origens. A análise de variância molecular (AMOVA) mostrou maior variação (75%) dentro dos grupos do que entre os grupos (25%). Esses resultados fornecem informações que podem ser utilizadas para desenvolver estratégias de conservação da fruta do dragão e programas de melhoramento para a obtenção de genótipos de pitahaya mais produtivos, com qualidade superior, alto rendimento e com resistência a fatores bióticos e abióticos.

Two chloroplast DNA (cp DNA) sequences and five inter-simple sequence repeat (ISSR) primers were used for amplification and sequencing. Chromas, Mega 7.0, DanSP5, and GenALEx were used to calibrate, splice, and analyze the sequence characteristics. PERMUT and PopGen 1.31 were used to analyze the genetic diversity parameters and genetic structure, and NTSYS was used to obtain the unweighted pair-group method with arithmetic means(UPGMA) clustering tree plot of 47 I. indigotica germplasm materials. ResultA total of 129 samples from 47 I. indigotica germplasm materials were successfully amplified and sequenced. The length of 2 cp DNA sequences after spliced was 1 412 bp, and there were 377 polymorphic variation loci, and 36 haplotypes. Fu and Li's D* test was significant (P<0.01). The values of Pi, HS, and HT based on cp DNA were 0.119 89, 0.787, and 0.891, respectively. The genetic differentiation coefficients of gene differentiation coefficient(Gst), nucleotide differentiation coefficient(Nst), and fixation index(Fst) were 0.117, 0.468, and 0.488, respectively, and the gene flow (Nm) was 0.615. The mean values of PPB, Shannon information diversity index(I), Nei's genetic diversity index(H), and Gst based on ISSR were 78.85%, 0.334 8, 0.218 6, and 0.754 4, respectively, and the Nm value was 0.162 8. ConclusionI. indigotica has high genetic diversity and abundant haplotypes at the species level, with abundant haplotypes. Genetic differentiation among different germplasm materials is obvious, and gene exchange is not frequent. Genetic variation mainly exists among populations. The population has accumulated various low-frequency gene mutations recently, suggesting that it has experienced significant regional expansion in the history.

ABSTRACT

Illumina Xten was employed for shallow sequencing of Panax ginseng(ginseng) samples, MISA for screening of SSR loci, and Primer 3 for primer design. Polymorphic primers were screened from 180 primers. From the successfully amplified polymorphic primers, 15 primers which featured clear peak shape, good polymorphism, and ease of statistics were selected and used to evaluate the genetic diversity and germplasm resources of 36 ginseng accessions with different fruit colors from Jilin province. The results showed that red-fruit ginseng population had high genetic diversity with the average number of alleles(N_a) of 1.031 and haploid genetic diversity(h) of 0.172. The neighbor-joining cluster analysis demonstrated that the germplasms of red-fruit and yellow-fruit ginseng populations were obviously intermixed, and pick-fruit ginseng germplasms clustered into a single clade. The results of STRUCTURE analysis showed high proportion of single genotype in pick-fruit ginseng germplasm and abundant genotypes in red-fruit and yellow-fruit ginseng germplasms with obvious germplasm mixing. AMOVA revealed that genetic variation occurred mainly within populations(62.00%, P<0.001), and rarely among populations(39%, P<0.001), but homogenization was obvious among different populations. In summary, pink-fruit ginseng population may contain rare genotypes, which is the basis for breeding of high-quality high-yield, and multi-resistance varieties, genetic improvement of varieties, and sustainable development and utilization of ginseng germplasm resources.

ABSTRACT

To clarify the content characteristics of mineral elements in different Artemisia argyi germplasm resources and their relationship with the quality properties of Artemisiae Argyi Folium, this study measured the content of 10 mineral elements including nitrogen(N), phosphorus(P), potassium(K), calcium(Ca), magnesium(Mg), aluminum(Al), manganese(Mn), iron(Fe), copper(Cu), and zinc(Zn) in 100 Artemisia argyi germplasm samples. Besides, their relationship with the quality properties of Artemisiae Argyi Folium was explored by correlation analysis, path analysis, and cluster analysis. The results demonstrated that the variation coefficient of the 10 mineral elements in Artemisiae Argyi Folium ranged from 12.23% to 64.38%, and the genetic diversity index from 0.97 to 3.09. The genetic diversities of N, P, and Zn were obvious. As revealed by the correlation analysis, N, P, and K showed strong positive correlations with each other. Except that Mg and Al were negatively correlated, Ca, Mg, Al, Mn, Fe, Cu, and Zn were positively correlated. The correlation analysis of mineral elements with the quality properties of Artemisiae Argyi Folium proved the significant correlations of 17 pairs of characters. According to the path analysis, P, K, Ca, and Mn greatly affected the yield of Artemisiae Argyi Folium, P, K, and Mg the output rate of moxa, N, P, and K the content of total volatile oil, P and K the content of eucalyptol, and P, K, and Ca the content of eupatilin. The 100 germplasm samples were clustered into three groups. Specifically, in cluster Ⅰ, the enrichment capacity of P, K, and Mg elements was strong, and the comprehensive properties of mineral elements were better, implying good development potential. Ca, Mn, Fe, and Zn elements in cluster Ⅱ and N and Al in cluster Ⅲ displayed strong enrichment capacities. This study has provided new ideas for resource evaluation and variety breeding of A. argyi and also reference for fertilizer application.

ABSTRACT

Cultivating salt-alkali tolerant rice varieties is one of the important ways to meet the increasing food demand of growing global population. In this study, twenty-one rice germplasms with different salt-alkali tolerance were treated with six salt-alkali concentrations at germination and seedling stages. The germination potential, germination rate, shoot length, root length, root number, fresh weight of shoot and seedlings were measured. The average value of salt damage rate was used to evaluate the salt-alkali tolerance. As the salt-alkali concentration increases, the inhibition on seed germination and growth became more obvious. Upon treatment with 1% NaCl plus 0.25% NaHCO3, the salt damage rate of germination rate has the largest variation, ranging from 0% to 89.80%. The salt damage rate of each trait shows a similar trend at all concentrations. Four germplasm resources with strong salt-alkali tolerance (Dajiugu, Nippobare, Mowanggu and 02428) and 7 sensitive germplasms were screened. The salt-tolerant gene sequence of 4 salt-alkali tolerant varieties and 3 sensitive germplasms were analyzed. OSHAL3 and OsRR22 were identical among the 7 germplasms, but SKC1 and DST showed clear variations between the salt-alkali tolerant and sensitive germplasms. Besides the salt-alkali tolerant germplasm resources, this study can also serve as a reference for mining of genes involved in salt-alkali tolerance and breeding of salt-alkali tolerant rice varieties.

RESUMEN La festuca alta (Festuca arundinacea Schreb.), está representada en la Argentina por poblaciones adaptadas del morfotipo continental, que son largamente persistentes. Es la principal especie forrajera perenne cultivada en la región templada en el país, productora de forraje en pastoreo directo. En este trabajo se presenta el desarrollo del mejoramiento genético y la contribución del mismo para el logro de una mayor productividad y mejor valor nutritivo con cultivares sintéticos modernos. Los caracteres considerados fueron: adaptación y persistencia en ambientes adversos, digestibilidad, flexibilidad de lámina de la hoja, y tolerancia a roya. Los cultivares más representativos de las etapas y criterios de selección considerados en este trabajo fueron: Pergamino El Palenque MAG, Palenque Plus INTA, Brava INTA, Baguala y Luján INTA.


ABSTRACT Tall fescue (Festuca arundinacea Schreb.) is represented in Argentina by adapted populations of the continental morphotype, which are long persistent. It is the main perennial forage species cultivated in the temperate region of the country, producing forage for extensive grazing. The development of fescue plant breeding and its contribution to the achievement of higher productivity and better nutritional value with modern synthetic cultivars was the aim of this project. The characters considered were: adaptation and persistence in adverse environments, digestibility, leaf softness and tolerance to rust. The most representative cultivars of the stages and selection criteria considered in this work were: Pergamino El Palenque MAG, Palenque Plus INTA, Brava INTA, Baguala and Luján INTA.

RESUMEN En el mejoramiento del tomate (Solanum lycopersicum L.) se ha logrado un incremento significativo para el rendimiento y otras características productivas en un período corto de tiempo. Como consecuencia se redujo notablemente la diversidad genética. Si bien el germoplasma silvestre se ha utilizado principalmente como fuente de genes de resistencia para enfermedades y plagas, nuestro grupo inició en la década de 1990, un programa de mejoramiento genético en tomate para mejorar la calidad del fruto con especial énfasis en incrementar la vida poscosecha y también ampliar la variabilidad genética con la incorporación de estos genes al gran cultivo. Hemos desarrollado diferentes poblaciones a partir del cruzamiento interespecífico entre el cultivar argentino Caimanta de S. lycopersicum y la accesión LA0722 de S. pimpinellifolium L. Mediante la generación de cruzamientos entre estos padres selectos y el posterior avance generacional de la selección se ha tratado de dilucidar las bases genéticas que definen la calidad del fruto. Para ello se integraron al programa de mejoramiento información obtenida de datos genómicos, posgenómicos y bioinformáticos. Al mismo tiempo hemos desarrollado cuatro nuevos cultivares con características de calidad de fruto superiores al ser comparados con híbridos comerciales. Para conservar y estudiar la diversidad del cultivo también estamos desarrollado una colección de germoplasma que en la actualidad cuenta con 162 genotipos de tomate de diferentes especies y orígenes. Además, se ha iniciado la transferencia directa de plantines a huertas urbanas y periurbanas para favorecer el acceso a semillas de estos cultivares desarrollados en instituciones públicas.


ABSTRACT The genetic improvement of tomato (Solanum lycopersicum L.) has achieved an increase for yield and other agronomic traits in a short period of time. As a consequence, genetic diversity has been notably reduced. Wild germplasm has been mostly used as a source of resistance genes for diseases and pests. Our group started in the 1990' a breeding program in tomato for improving fruit quality, with special emphasis on increasing fruit shelf life and broadening the genetic variability with the incorporation of wild genes. We have developed different populations from the interspecific cross between the Argentine cultivar Caimanta of S. lycopersicum and the accession LA0722 of S. pimpinellifolium L. Through crosses between these selected parents and the subsequent generational selection advance, we attempted to elucidate the genetic bases that underlie tomato fruit quality. To do that, we use state-of-the-art technology available in the field of genetics and breeding programs, including genomic, post-genomic and bioinformatic data. At the same time, we have developed four new cultivars with improved fruit quality traits compared to commercial hybrids. To conserve and study the tomato diversity, we have developed a germplasm collection that currently contains 162 tomato genotypes from different species and origins. In addition, we have started a direct transfer of our cultivars to urban and peri-urban community orchards to facilitate them the access to genotypes that were developed in Argentine public institutions.

Varronia curassavica has anti-inflammatory properties because of the terpenes, α-humulene and ß-caryophyllene, present in the essential oil. The objective of this study was to evaluate the influence of seasonality on the essential oil of V. curassavica accessions. Leaves from six accessions were collected from the Germplasm Bank of the Federal University of Minas Gerais over 12 months. Correlations between the essential oil content and meteorological factors were determined. Gas chromatography analysis coupled with mass spectrometry was conducted to determine the chemical composition of the essential oil. The content and chemical composition of the oil varied throughout the year. Relative humidity was correlated with accessions ICA-VC2 (-0.64) and ICA-VC4 (0.68). ß-bourbonene, ß-elemene, spathulenol, germacrene, caryophyllene oxide, α-humulene, and ß-caryophyllene occurred in all accessions. Accession ICA-VC3 exhibited lower variation (22.17%), higher average (0.97%) essential oil, and maintained an average abundance of α-humulene greater than 2.6%, which is the amount necessary for phytotherapeutics.


Varronia curassavica tiene propiedades antiinflamatorias debido a los terpenos, α-humuleno y ß-cariofileno, presentes en el aceite esencial. El objetivo de este estudio fue evaluar la influencia de la estacionalidad en el aceite esencial de las accesiones de V. curassavica. Se recolectaron hojas de seis accesiones del Banco de Germoplasma de la Universidad Federal de Minas Gerais durante 12 meses. Se determinaron las correlaciones entre el contenido de aceite esencial y los factores meteorológicos. Se realizó un análisis de cromatografía de gases junto con espectrometría de masas para determinar la composición química del aceite esencial. El contenido y la composición química del aceite varió a lo largo del año. La humedad relativa se correlacionó con las accesiones ICA-VC2 (-0,64) e ICA-VC4 (0,68). En todas las accesiones aparecieron ß-bourboneno, ß-elemeno, espatulenol, germacreno, óxido de cariofileno, α-humuleno y ß-cariofileno. La accesión ICA-VC3 mostró una menor variación (22,17%), un promedio más alto (0,97%) de aceite esencial y mantuvo una abundancia media de α-humuleno superior al 2,6%, que es la cantidad necesaria para los fitoterápicos.

ABSTRACT

ABSTRACT Plant genetic resources for food and agriculture are ex situ conserved in germplasm banks as samples (accessions) of natural or naturalized populations, either as the originally sampled propagules (mainly seeds) or their multiplications. The premises underlying ex situ conservation are that (a) it is the safest and cheapest alternative for germplasm preservation for future generations and (b) accessions are representative of the genetic diversity encountered in nature. In the past decades, ideas, alternatives and considerations have been put forward on the topic, and protocols have been devised for plant germplasm sampling, conservation and multiplication. However, limitations in the management efficiency of germplasm banks have been pointed out by international organizations. In our opinion, germplasm banks in general need to revise their functioning and management at the light of principles and methods of Genetics. To that end, it is necessary to consider the reproductive biology of higher plants -whose genetic consequences at both the individual plant and the population levels are not always either fully understood or taken into account in devising the protocols-, the genetic structures of wild and cultivated populations, and the course of the genetic material in the populations. In this paper, we discuss the three topics and provide an example of a national forage breeding program, from germplasm bank accessions as the germplasm of origin to the obtainment of commercial cultivars. Finally, we present a proposal as a base for discussion among curators, researchers and breeders.


RESUMEN Los recursos genéticos vegetales para la alimentación y la agricultura se conservan ex situ en bancos de germoplasma como muestras (introducciones) de poblaciones naturales o naturalizadas ya sea como propágulos originales (mayoritariamente semillas) o sus multiplicaciones. Las premisas subyacentes son que (a) es la alternativa más segura y barata de preservación de germoplasma para futuras generaciones y (b) las introducciones son representativas de la diversidad genética que se encuentra en la naturaleza En las últimas décadas, se han presentado ideas, alternativas y consideraciones sobre el tema y se han elaborado protocolos para el muestreo, conservación y multiplicación de germoplasma. Sin embargo, organizaciones internacionales han señalado limitaciones en la eficiencia del manejo de los bancos de germoplasma. En nuestra opinión, se necesita revisar el funcionamiento y manejo de dichos bancos en general a la luz de los principios y métodos de Genética. Para tal fin, es necesario considerar la biología reproductiva de las plantas superiores -cuyas consecuencias genéticas a nivel de planta individual y de población no se comprenden en su totalidad o no se consideran al idear los protocolos -, las estructuras genéticas de poblaciones naturales y cultivadas, y el curso del material genético en las poblaciones. En este trabajo discutimos los tres temas y proveemos un ejemplo de un programa nacional de mejoramiento de forrajeras, desde las introducciones como germoplasma de origen hasta la obtención de cultivares comerciales. Finalmente, presentamos una propuesta como base de discusión entre curadores, investigadores y mejoradores.
